Multiplex Assays for Cellular Signaling Research

The Bio-Plex phosphoprotein detection product line collectively refers to Bio-Plex phosphoprotein assays and Bio-Plex total target assays. These assays make it possible to simultaneously monitor multiple phosphorylation events with minimal sample volume requirements and minimal hands-on time. Whether you're evaluating the phosphorylation level of specific phosphoproteins or the expression level of specific target proteins, it's quick and easy to get the data you're looking for.

Bio-Plex Phosphoprotein Assays

Bio-Plex phosphoprotein assays are bead-based multiplexable assays that directly detect phosphorylated proteins in lysates derived from cell culture or tissue samples. Using these 96-well filter-plate format assays, you can profile the specific phosphorylation state of multiple proteins with as little as 25 µl of lysate. Choose from a broad assay selection and use the assays individually to analyze samples for one target at a time, or combine them to create a multiplex assay to test for a specific set of phosphoproteins in a single sample. Custom-mixed multiplex assays are available through Bio-Rad's online x-Plex assay service.

Bio-Plex phosphoprotein assays feature:

  • Highly specific antibodies exclusively developed and validated by Cell Signaling Technology, Inc.
  • A 96-well filter-plate format that yields significantly higher throughput than traditional methods
  • Higher-specificity sandwich format that minimizes cross-reaction with other cellular proteins
  • Direct measurement of kinases that have been activated within the cell
  • Minimal hands-on time and sample volume requirements; for assay workflow, click here

Bio-Plex Total Target Assays

The selection of Bio-Plex assays includes Bio-Plex total target assays that measure specific phosphoprotein levels, regardless of phosphorylation state, in cell culture or tissue sample lysates. While the Bio-Plex total target assay reports the abundance of the target protein in one well, the Bio-Plex phosphoprotein assay reports the level of phosphorylation of the protein in a separate well. Bio-Plex total target assays share the same format, protocol, and features of Bio-Plex phosphoprotein assays.

* EGFR and S6 ribosomal protein assays are not multiplexable; they are available only as singleplex assays. The remaining phosphoprotein assays are multiplexable; however, not all multiplex combinations are recommended. How to Order

Bio-Plex phosphoprotein and total target assays and associated reagents are designed to provide up to 96 data points for each assay. Two packaging options are available: one 96-well plate and ten 96-well plates. For phosphoprotein detection, you will need to order the following components:

Available as Individual, Premixed, or x-Plex Assays

Individual (singleplex) phosphoprotein and total target assays are designed to provide more flexibility. These assays can be used independently or combined to test for a specific set of proteins in a single sample. Premixed assays are preconfigured multiplex assays; they can be combined with other kit formats to customize your multiplex combination. x-Plex assays are custom-mixed multiplex assays that are quality-tested at Bio-Rad and are a convenient format for repeat sample testing. Click here to obtain catalog numbers online for one 96-well plate format and ten 96-well plate format x-Plex assays. Once the x-Plex assay catalog number has been identified, orders may be placed as with any other Bio-Rad product.
